My existing application has multiple forms using Syncfusion controls. Will I have to modify and save every form to ensure proper licensing for deployment?

No. Licensing is for a whole application and is per product. As suggested in licx.htm, you can ensure that a proper licenses.licx file is created by touching/modifying a form and then saving it.

If you have an application that has multiple forms using various Syncfusion controls, you could for example add a dummy form to your project and then drag-and-drop controls and then save the form so that a proper licenses.licx file is created and then delete the dummy form. For example, if you have an application with multiple forms which uses say Essential Tools controls along with Essential Grid and Essential Edit, all you would need to do is add a form and drop one Tools control (that you are using) along with Grid and Edit for this application to be licensed properly for deployment.
